Trypsin has been used in a study to assess the potential application in animal cell culture of an alkaline protease from a non-toxigenic mangrove isolate of Vibrio sp. V26.[1] Trypsin has also been used in a study to improve the detection of fungi in eosinophilic mucin.[2]
 

生化/生理作用

该抑制剂能够在较小程度上抑制胰蛋白酶、胰凝乳蛋白酶和纤溶酶。它还能抑制蛋白酶血浆激肽释放酶和凝血因子X，机制与抑制胰蛋白酶类似。胰蛋白酶抑制剂不会抑制金属蛋白酶、基于组织的激肽释放酶、酸性蛋白酶或硫代蛋白酶。该抑制剂通过与蛋白酶活性位点按1:1化学计量形成复合物，然后在抑制剂上切割单个精氨酸-异亮氨酸键。抑制作用是可逆的且具有pH依赖性。

The soybean trypsin inhibitor is a monomeric protein containing 181 amino acid residues in a single polypeptide chain crosslinked by two disulfide bridges.

一个胰蛋白酶单位是指在25°C、pH 7.6条件下，以N-α-苯甲酰基-L-精氨酸乙酯（BAEE）为底物，每分钟产生0.001的 A253所需的酶量。
One trypsin unit will produce a ΔA253 of 0.001 per min with BAEE as substrate at pH 7.6 at 25 °C; reaction volume 3.2 ml, 1 cm light path.
 

制备说明

胰蛋白酶抑制剂能够以10 mg/mL溶于水和磷酸盐缓冲液。它能够以1 mg/mL溶于平衡盐溶液和无血清培养基中。浓度大于10 mg/mL的浓缩液可能浑浊，呈黄色至琥珀色。使用胰蛋白酶消化细胞后，重悬于胰蛋白酶抑制剂溶液中，使得用于解离的胰蛋白酶溶液浓度达到1 mg/mL。然后将细胞悬浮液以1000rpm离心，得到细胞沉淀。

在2-8℃下短期储存时，溶液可保持活性。溶液分装后可在-20℃冷冻并保持稳定。
